Specimen Volume: 5 mL of serum
Collection Instructions: Allow specimen to clot for 30 minutes. Spin specimen in centrifuge.
Centrifuge specimens and remove the serum or plasma from the cellular material within 4 hours of collection.
Colorimetric
The VITROS SALI Slide is a multilayered, analytical element coated on a polyester support. A drop of patient sample is deposited on the slide and is evenly distributed by the spreading layer to the underlying layers. Salicylate in the sample is oxidatively decarboxylated to catechol. Catechol is oxidized by tyrosinase to o-quinone, which couples with MBTH to produce a dye. The amount of dye is proportional to the concentration of salicylate in the sample. The intensity of the dye is measured by reflectance spectrophotometry.
Salicylate belongs to a class of analgesic, antipyretic, and anti-inflammatory drugs that includes aspirin. Salicylate measurements are used in the diagnosis of salicylate overdose and in monitoring salicylate therapy.
