The VITROS UPRO Slide is a multilayered, analytical element coated on a polyester support. A drop of patient sample is deposited on the slide and is evenly distributed by the spreading layer to the underlying layers. The binding of protein to a pyrocatechol violet-molybdate complex in the presence of oxalate causes a shift of the absorption maximum from 450 to 670 nm. After a fixed incubation period, the reflection density of the dye is measured spectrophotometrically. The amount of colored complex is proportional to the protein concentration in the sample.
